转录组测序的研究对象为特定细胞在某一功能状态下所能转录出来的所有mRNA。转录组测序能够从整体水平研究基因表达量以及基因结构,揭示特定生物学过程中的分子机理;目前已广泛应用于基础研究、临床诊断、药物研发和分子育种等领域。
原核链特异性转录组 是研究原核生物基因表达状况的一种有效手段。相对于传统转录组测序而言,链特异性转录组可以确定转录本是来自正义还是反义DNA链,更能准确地统计转录本的数量和确定基因的结构,同时可以发现更多的反义转录本。目前该技术被广泛地应用于原核生物的基因结构和基因表达调控等研究领域。
原核生物链特异性转录组项目,可以挖掘新的ncRNA与反义(antisense)转录本;还可以进行基因结构的研究,例如操纵子鉴定,为基因网络调控的研究提供有力支持。同时探究细菌在不同时空条件下的基因表达变化,揭示基因组结构信息,深入挖掘sRNA调控信息。
