DNA甲基化是重要的表观遗传学标记信息,在维持正常细胞功能、遗传印记、胚胎发育及人类肿瘤发生中起着重要作用。获得全基因组范围内所有甲基化位点及水平数据,对表观遗传学的时空特异性研究具有重要意义。全基因组甲基化测序(Whole Genome Bisulfite Sequencing, WGBS)技术,采用亚硫酸氢盐处理基因组DNA,使未发生甲基化的胞嘧啶(C)被转化为尿嘧啶(U),而甲基化的胞嘧啶不变。亚硫酸氢盐处理后的DNA进行全基因组测序,并与参考基因组进行比对,从基因组水平实现单碱基分辨率的、高精确度甲基化水平分析,广泛应用于细胞分化、组织发育、动植物育种及人类健康与疾病研究等领域。
